Focal points-adhesive nano-pillar array for human mesenchymal stem cell (hMSC) |

Cell differentiation can be controlled by manipulating focal adhesion point because cell attachment is related to cell behaviours. In order to induce differentiation of hMSCs, we prepared nano-scale pillar array modified with focal adhesion protein. The master with nanoscale-hole arrays was patterned on silicon wafer by e-beam lithography. Nano-pillar array was prepared using poly (dimethylsiloxane) (PDMS) against the master. PDMS replica was placed on a fibronectin (0.1mg/ml) coated slide and applied light pressure. After fibronectin coating, hMSCs (5x103cells/device) were cultivated for 24h. To measure the expressions of focal adhesion proteins, immunofluorescence was performed in hMSCs. The nano-pillar array was successfully fabricated at spacing of 3, 5, 10 μm with diameter of 500nm and height of 1μm on PDMS replica. Focal adhesion proteins were clearly visualized according to the spacing gap between pillars on the PDMS. Filopodias of hMSC stretched out along the top of pillars. |
