Branched polyethyleneimine(PEI) was well known as high gene transfection agent. However, utilization of PEI as gene carrier was limited because of high cytotoxicity. To solve this problem, PEI was introduced to chitosan(CS) back bone by coupling agent. Structural analysis of chitosan-grafted PEI(CSPEI) was confirmed by 1H-NMR and synthesized successfully. In addition, binary complex of pDNA/CSPEI was prepared by electrostatic charge interaction. its gelretardation assay showed that pDNA was completely condensed by CSPEI. Moreover, it can protect from DNase and release by heparin with negative charge. Also, transfection assay showed that CSPEI was effectively tranfected more than PEI. It means that cytotoxicity of PEI was reduced by introduced chitosan. These results suggest that CSPEI can be applied as gene carrier.
