Quantitative analysis strategy for N-acyl homoserine lactones (AHLs) as quorum sensing modulators via MALDI-TOF MS |
초록 |
As N-Acyl homoserine lactones (AHLs) are used for antibacterial drug development and cell-to-cell communication, these are important secondary metabolites. Here, we proposed a highly sensitive and reliable MALDI-MS based 3-oxo AHL quantitation method, which ketone group of AHLs were derivatized with Girard’s reagent T to (GT) to produce a permanent cationic charge state [M]+. By the chemical derivatization of 3-oxo AHLs, its detection sensitivity was improved and the derivatization also allowed accurate quantitative analysis from the PAO1 culture supernatants. This method could be applied for developing high-throughput and sensitive detection methods of quorum sensing signal molecules. |
