제목 Development of riboswitch-based sensor devices that specifically respond to intracellular concentration of naringenin through in vitro and in vivo selection
초록 Measuring Intracellular concentration of metabolites enables facile control of the pathway gene expression and screening or selection for improved producer strains. Transcription factor-based sensors are often accompanied by off-target effect and delayed response which result from trans-regulatory mechanism. On the contrary, riboswitch could bypass the aforementioned problems. Here we demonstrate the process for developing riboswitches which specifically sense naringenin by modulating concentration of naringenin during enrichments and the sequence of negative and positive selections. We found that selected riboswitches work at useful operational ranges which are dependent on the screening condition applied to the library. Finally, we validated the specificity of the riboswitches for naringenin against other flavonoids. We expect that these new riboswitches could be applied to diverse applications.
