In our previous works, we confirmed that a PLCL(Poly[L-lactide-co-ε-caprolactone]) scaffold fabricated by scCO2 has better ability than a scaffold fabricated by solvent casting method for cartilage tissue engineering. However, we could not fabricate PLCL scaffolds at 35℃ because PLCL polymer has high molecular weight and linear part in the chemical structure. In this study, we introduced cosolvent-scCO2 foaming system to fabricate a TGF-beta3 encapsulated PLCL scaffold at 35℃. Morphologies of the scaffold was observed by FE-SEM and porosity was measured by mercury intrusion porosimeter. TGF-beta3 molecules were homogeneously contained in the scaffold and released out more than 8 weeks. And also, we confirmed the chondrogenic ability of the scaffold from in vivo tests. From the results, we considered that the PLCL scaffold fabricated by cosolvent-scCO2 foaming system could successfully be used for cartilage tissue engineering. |
