Monitoring of polymer carrier/plasmidDNA (pDNA) dissociation is important to understand the mechanisms of polymeric gene delivery system for a rational design of vector. We monitored the carrier/pDNA dissociation using carbon dot (CD) which received attention for promising alternative of quantum dot (QD). The fluorescence of CD is quenched by Au nanoparticle when they interact with non-labeled pDNA since CD and Au surface is modified with highly cationic polymer, polyethylenimine (PEI). When the pDNA were dissociated from the complex, the fluorescence displacement occurred by the distance increment between CD and Au. Also, the CD/Au carrier showed not only great transfection efficiency but also high cell viability at the optimum condition. Overall, this CD/Au carrier system were found to be a great gene delivery carrier and effective at monitoring of carrier/pDNA dissociation in a non-labeled manner, thus great potential for studying the mechanisms of polymeric pDNA delivery system. |
