ω -hydroxy fatty acid is are multifunctional precursor of many polymers such as pseudo-ceramide. E.coli has been widely used for the production of ω -hydroxy fatty acid. Here, to produce ω-hydroxy lauric acid from lauric acid, monooxygenase CYP153A33 which is origined from Marinobacter Aquaeolei was cloning to E.coli. It is already known that CYP153A33 is good for ω -hydroxylation of dodecanoic acid. We have engineered substrate binding site of P450 to make higher substrate binding efficiency. We found 3 points in substrate binding site by docking simulation, and checked activity by whole cell biotransformation. By this in vivo study, we found that CYP153A33(E142A) has higher activity than wild type. After optimized P450 reaction system, finally we can make 3.46g/L of 12-hydroxy dodecanoic acid from 4g/L of dodecanoic acid. |
