Nucleotides (NTs) such as ATP, GTP, CTP and TTP are very important molecules as signal molecules, DNA components, cofactors, and energy sources in the body. Among them, especially, ATP (adenosine triphosphate) is the most essential bioenergy for maintaining life. However, its susceptible hydrolysis could lose phosphate group from ATP and then ATP could weaken its bioenergetic power. Thus, we synthesized polynucleotides (pNTs) using EDC coupling reaction. The synthesized pNTs were water-soluble and high molecular weights (∼ 500 kDa). Also, pATP exhibited low cytotoxicity (IC50; 2 mg/mL). In a energy-deprived condition (a low temperature, 4 oC), pATP as an bioenergy source increased cell viability up to 20% than the cell control without pATP treatment. In addition, pATP prevented low temperature-induced reduction of bPEI-based transfection efficiency. Based on the results, pNTs as a bioenergy source could protect cell survival rates from energy deprivation-mediated cellular damages. |
