Screening phage-displayed peptide libraries is a useful tool for the identification of peptides that selectively bind to biomarkers or cancer cells. To identify peptides that sense myoglobin, one of key biomarkers of myocardial infarction, we screened peptide libraries for peptides that bind to myoglobin. Two peptides, MGB-1 and MGB-2, showed selective binding to coated myoglobin compared to albumin. Using the peptides conjugated with magnetic beads or biotin, we are currently developing a Sandwich ELISA for myoglobin. In addition, we have identified Bld-1 peptide that binds to bladder tumor cells. To sense bladder tumor cells in the urine, cells from urine were stained with Pap and Bld-1-labeled magnetic beads, which could be detected as shiny, gold-colored beads. The results suggest that the combination of Pap staining and Bld-1-based sensing of cells can improve the sensitivity and specificity of bladder cancer diagnosis. |
