Conjugated polyelectrolytes as an optical platform for fluorescence-based DNA detection |

One successful in-situ DNA detection was demonstrated by employing fluorescence resonance energy transfer (FRET) from cationic conjugated polyelectrolytes (CCPs) to fluorophore-labeled peptide nucleic acid (PNA) or DNA (hybridized with the complementary target DNA), in which electrostatic interactions between CCPs and negatively charged DNA bring them into close proximity to satisfy FRET distance condition for strand-specific DNA detection. This presentation reports the several approaches how to control the fine-structure of the electrostatic complex (CCPs/DNA) and competition between energy harvesting FRET and energy-wasting photo-induced charge transfer, for maximizing the FRET-induced detection signal. Simple and general strategies will be discussed for the optimized conjugated polymer-based FRET biosensors. |
