Pretreatment Step in Separation Process for Taxol
초록
항암제로서 탁월한 효능이 있는 taxol을 주목나무에서 용매를 사용하여 추출하고 분배과정으로 정제하였다. 추출에 사용된 용매는 methanol이었으며, 추출액은 물과 methylene chloride(MC)의 분배과정을 거쳐서 농축하였다. 농축액에 잔존하는 lipid, chlorophyl, pigment 등의 불순물을 제거하기 위해서 전처리 단계에서 open column을 사용하였다. Silica를 충진시킨 column에 추출액 시료 10 ml를 이동상과 함께 통과시켜 용출액을 색층(color band)별로 분취하였다. 실험에 사용된 이동상은 hexane, methylene chloride, chloroform, acetone, methanol, ethanol, IPA, 1-propanol을 단일 성분 또는 2성분계로 조합하여 실험하였다. 실험결과에 의하면 MC/acetone의 이성분계 이동상에서 색층이 가장 선명하게 구분되었으며 이동상의 조성을 97/3에서 75/25(vol.%)의 구배용매 조성법에서 용출시간이 가장 빨랐다.
Taxol, a powerful anti-cancer agent, was extracted and purified from yew tree by solvents. The solvent used in extraction was methanol, and the extracts was further concentrated by the partition of water and methylene chloride. Such impurities as lipid, chlorophyl, pigment, et al. were removed in the pretreatment step with open column, where silica packings were packed. The extracts 10 ml passed through the column with mobile phase, and the effluents were collected by the bands developed in the column. The solvents used were hexane, methylene chloride, chloroform, acetone, methanol, ethanol, IPA, and 1-propanol. They were used as mobile phase with a single or binary combination. From the experimental results, the mobile phase of MC/acetone most clearly showed the developed band of the extract, and the elution time was fasted by the gradient mode(75/25 after 97/3 vol.% of MC/acetone).
